Bioavailability of Drugs, Drug Candidates, Dietary Chemicals, Nutrients, and Micronutrients, Caco-2 Cells, Intestinal Perfusion, Microsomes, Drug Metabolizing Organs, In Vivo Pharmacokinetic Studies, Oral Bioavailability
My laboratory is devoted to study the factors that govern the bioavailability of drugs, drug candidates, dietary chemicals, nutrients and micronutrients. To achieve the proposed goal, we deploy a variety of biological model systems including Caco-2 cells, intestinal perfusion, microsomes and S9 fractions that are enriched in metabolic enzymes from various drug metabolizing organs. We also conducted in vivo pharmacokinetic studies. We also have all the necessary state-of-the-art analytical instruments available for conducting these studies, including two LC-MS/MS with one of the most sensitive instruments (perhaps the most sensitive) in AB Sciex 5500 QTrap. A major undertaking of our lab is to engage in the study of glucuronidation of flavonoids and phenolics for the purpose of improving their oral bioavailability. Toward this goal, we have conducted in vitro study using cellular and subcellular models (e.g., microsomes), established new cell lines, and characterized the pharmacokinetic profiles of flavonoids in normal and efflux transporter (e.g., Bcrp or UGt1A) knockout mice/rats. We have shown that inhibition of efflux transporters could improve the bioavailability of flavonoids. Our lab is also interested in biopharmaceutical characterization of natural compounds active in disease prevention and treatment. Recently, we have been active in understanding how to characterize herbal formulations (e.g., ginseng) biopharmaceutically so that we can prepare more active fractions and establish quality control and quality assurance criteria that are based on sciences. We have and will continue to collaborate with cancer biologists and other specialists to determine the exposure of relevant agents in vivo and develop means to improve the bioavailability. These types of studies are very important in the preclinical characterization of agents, and will facilitate their transition to translational studies in humans.
